About Som Chatterjee

Staphylococcus aureus is one of the most adaptable pathogens that can cause severe and lethal infections, complicating treatment especially since there are no available vaccines. During my postdoctoral training I worked on Phenol Soluble Modulins (PSMs), a group of cytolytic toxins secreted by S. aureus. I discovered the PSMs transporter in S. aureus, its mechanism of regulation and function. Currently my work focuses on beta-lactam resistance in the bacteria. Beta-lactam drugs are antibiotics of choice for treatment of S. aureus infections due to their excellent efficacy and safety. They are potent inhibitors of the bacterial cell wall synthesizing enzymes, penicillin binding proteins (PBPs). However, broad spectrum beta-lactam resistance, i.e. methicillin resistance, is common in S. aureus and is typically mediated by PBP2a, which is encoded by the gene mecA. We have identified an uncanonical (non-mecA mediated), yet equally potent mechanism of beta-lactam resistance in S. aureus. Currently the lab is working to decipher the mechanistic basis of this resistance.

I joined NIH in 2007 and became interested in the mechanisms used by S. aureus for secretion of phenol-soluble modulins (PSMs), a group of cytolytic peptide toxins that are key virulence determinants of Staphylococcus species. My work led to discovery of the PSMs transporter (Pmt), an orphan ABC transporter that we showed was critical for staphylococcal growth and virulence. This work was recently published in Nature Medicine and indicates the importance of Pmt as a potential therapeutic target. Current work in the lab is exploring Pmt as a candidate vaccine antigen against Staphylococcus infections.
