    Projects Undertaken:• “Transgenic approaches for Frost Resistance in Indian Mustard.” Evaluated transgenic Brassica juncea plants for their capacity to withstand frost stress. Cloned known positive and negative regulators of cold stress pathway under the control of either embryo-specific Napin promoter or stress-inducible RD29A promoter to introduce frost tolerance in B. juncea. Standardized conditions of radiative frosting, in a frost chamber, to mimic frosting in fields and also optimized stress regimes to induce lethality in untransformed plants. Tested untransformed and transformed B. juncea lines for frost tolerance, under standardized stress regimes, which resulted in few plants showing high frost tolerance as compared to the wild type • “Transcriptome profiling of Brassica juncea siliques during cold stress andvarious developmental stages.”• “Isolation of cold-inducible embryo-specific promoter.” Used genome walking approach to identify and clone a cold-inducible and embryo-specific promoter from B. juncea. • “Unravel the dynamics of chromatin architecture in response to heat, salt, drought and cold stresses in Oryza sativa by using two cross validating methodologies [DNaseI hypersensitive assay (DNase) and Formaldehyde Assisted Isolation of Regulatory Elements (FAIRE) coupled with high throughput sequencing]”Further superimposed the transcriptome information on the identified open chromatin regions (OCRs), to decipher importance of various cis-elements in regulating gene expression.• “Mutant analysis and open chromatin study during abiotic stresses in Arabidopsis.”
