ResearchColorado State University Aug 2011 - Apr 2012Fort Collins, Co, UsStem Cell Cryopreservation Protocol Revisited: A comparison of current cryopreservation of stem cells to a proposed technique incorporating the addition of cholesterol and propylene glycolTested the effects of the cryoprotectant propylene glycol (PG), membrane protecting cholesterol-loaded β-cyclodextrin (CLCs), and trehalose on the cryosurvival of stem cells. Previous studies reported PG was superior to DMSO for cryopreserving mouse stem cells. Therefore, I hypothesized PG would be more effective than DMSO when freezing three different stem cell lines. Other studies testing the effects of CLCs on Chinese Hamster Ovary cells and bull sperm led to my hypothesis that increasing membrane cholesterol content would result in higher cryosurvival rate. The stem cells tested were uncryopreserved horse mesenchymal stem cells and previously cryopreserved human and mouse trophoblast stem cells. The control treatment was the standard 10% DMSO while the experimental groups were frozen in 10% DMSO + CLC, 7.35% PG, and 7.35% PG + CLC. Cells were cooled to -41°C at -1°C per minute and stored in liquid nitrogen for 1-3 weeks. Cryosurvival rates were determined using flow cytometry. DMSO-treated cells survived freezing more effectively than PG for all three stem cell lines. Interestingly, the previously cryopreserved BeWo human trophoblast stem cells showed an increase in cell viability with the addition of CLCs with both DMSO and PG. Although the results of the horse and mouse stem cell experiments indicate that CLC and PG do not increase stem cell viability after cryopreservation, the increase in BeWo cryosurvival shows that future work using different CLC and cryoprotectant concentrations as well as different cryoprotectants, cell types, and cryomedia could potentially yield improved stem cell viability after cryopreservation. -
