Blood processing, sterile sampling and eventual testing of processed blood components to perform various studies designed to compare, validate, or test the quality of related products (Leucocyte filters, blood bags, additive solutions etc) manufactured onsite or in adjunct facilities. • Processing of whole blood into its components using either a hard spin centrifugation (plasma and red cells) or a soft spin centrifugation (plasma, red cells and buffy coat).• Pooling and splitting of whole blood, red cells or plasma into 2, 3 or 6 equal parts.• Separation of the spun components using various types of expressers (manual, Optipress II, Compomat G-4). • Sterile sampling of the components using sample diversion pouches• Filtering (across Leucocyte reduction filters) of the processed red cells removing any WBCS.• Analysis of various ion concentrations, pH, blood gases and cell counts using the Bayer Rapidlab 1265 Blood Gas Analyzer• Use of the Cell Dyn 3200 to measure WBCs, Platelets, and blood gases in blood samples.• Extraction of intracellular metabolites using Percholoric acid.• Evaluation of ATP, 2,3 DPG and plasma hemoglobin concentrations using the BioTek Microplate Reader• Staining red cell and platelet samples with propidium Iodide to account for resdiual WBC’s and platelets concentration using flow cytometry• Maintenance, calibration and running of QC controls for the above mentioned equipment on a daily basis.• Preparation of various chemical solutions used in a wide range of assays. • Analysis, statistical interpretation and subsequent data entry of results obtained from assays.
