Over the past 12 years my career has been primarily focused on Neuroscience in the disciplines of genetics and molecular/cellular biology and applying these to a range of human neurodegenerative disease; including Multiple sclerosis (MS), Motor Neuron Disease (MND), Parkinson’s and a range of eye diseases.My current role within the Neurogenetics laboratory at The Florey Institute has been focused on developing and optimising lab techniques to enable our single cell genomics research program focused on MS and MND. I have also overseen the establishment of our neuropathological tissue biobank and developed lab protocols for fluorescence-activated nuclei sorting (FANS) of different cell types within the brain. Another important role I have, is to manage multiple different biobanks, these include; the MS blood DNA biobank, MS brain tissue biobank and MND brain and spinal cord tissue biobank, all held at the Florey Institute. Overall we are the custodian of over 9500 patient DNA samples and close to 1000 tissue samples. Clinicians are still recruiting and adding patients to this biobank. Currently our lab is also involved in an exciting opiod pharmacogenomics project in collaboration at the VCCC. Prior to my current role, I held positions at the Centre for Eye Research (Melbourne), here I built upon my skills and knowledge of working with primary iPSC’s and a developed more specialised clinical approach to research. The iPSC projects I worked on involved in-patient specific mutation correction using CRISPR genetic manipulation techniques. Other research I have been involved in has investigated the role of the MYRF transcription factor in the maturation of oligodendrocyte precursor cells into fully differentiated myelinating oligodendrocytes. We were able to show that MYRF is a membrane-associated transcription factor with a highly complex structure that autoproteolytically cleaves to directly activate myelin genes within the brain.
