During my doctoral time I characterized the P. aeruginosa YfiR protein in conjunction with its interaction partner YfiN, a protein located in the inner membrane of the cell. Structural analysis of YfiR revealed a dimeric assembly of the protein in the crystal. SEC-MALLS analysis of wildtype YfiR and a dimeric-interface mutant demonstrated that the protein is dimerizing with a high affinity in solution, via the same interface observed in the crystal structure. In a next step, a functional assay was established using YfiN and YfiR, which provided evidence that YfiR is inhibiting YfiN activity in-vitro.Ultimately, my thesis provided solid biochemical foundation for future work towards an ever more detailed understanding of the YfiBNR system of P. aeruginosa, which thereby provided insight into bacterial signaling.Thesis title: ‘Structural and functional investigations of the Pseudomonas aeruginosa YfiBNR-system’Technical expertise:- Structure determination using X-ray crystallography-Upstream Processing (USP) including construct design and expression using bacterial cells- DSP: Process design and optimisation to purify the recombinant (membrane) proteins (HPLC, FPLC, SEC, IEC....)- Analytical Method Development and Optimisation in order to characterise Biologics- Crystallization of proteins using high throughput methodsBiophysics- Multi-angle-laser-light scattering (DLS/MALLS/SEC-MALLS)- Thermal shift assay, Differential scanning calorimetry- Isothermal calorimetry titration (ITC)Biochemistry- PCR- protein characterization (SDS-PAGE, Western Blot)- Experience in handling of membrane proteins
