Worked on In Vitro Generation of Dendritic Cells from Hematopoietic Stem Cells: Characterisation, Cryopreservation and Evaluation of their role in cancer immunotherapy.Applied tissue culture techniques, molecular biology techniques, biochemical techniques in this research project. Generated dendritic cells on large scale from cord blood HSCs & studied phenotypic & functional characterization of in vitro generated Dendritic Cells.Studied antigen presentation to T Cells, generation of Cytotoxic T lymphocytes and assessment of anti tumor activity in vitro.Studied cryopreservation comprising of generation of DC’s from frozen cord blood units and freezing & revival of in vitro generated DC’s. Tested effect of polyunsaturated fatty acid - arachidonic acid, as supplement in the differentiation media for in vitro DC generation, improving functional properties of DCs.Studied mechanistic basis behind improvement of functional properties of DCs generated with and without arachidonic acid through transcript levels of three enzymes associated with arachidonic acid pathway, such as COX-1, COX-2, PLA2 by RT-PCR.Studied homing ability of the in vitro- generated cells in NOD/SCID mice model. The research work concludes that these DCs may be an attractive source of allogeneic DCs in future immunotherapy trials
