After my Master degree achieved at the Louis Pasteur University of Strasbourg, I have joined the Molecular and Cellular Oncology Laboratory at the CRP-Santé (Luxembourg) in April 2008 in order to start my PhD. In the first two years, one aspect of my project consisted in the analysis of the plant actin cytoskeleton modifications caused by mechanical cues. I particularly focus on the potential roles of the actin-bundling proteins called LIM proteins. Later, I shifted my attention towards another important family of actin-binding proteins: the actin-depolymerizing factors (ADFs). ADFs play key roles in many cellular processes in both plants and animals and are involved in several diseases. Interestingly, plants possess particularly diversified ADF families with 11 functional isovariants in Arabidopsis thaliana. Most ADFs characterized so far function through severing actin filaments and/or accelerating pointed-end depolymerization. By characterizing and comparing the actin-regulatory activities of several Arabidopsis ADFs, I found that two of those displayed atypical activities. Indeed, both ADF9 and ADF5 (which together form the subclass III) exhibit a surprisingly high ability to stabilize and crosslink actin filaments into long and thick actin bundles suggesting an additional degree of complexity in the comprehension of the biological functions of the ADF family. My thesis defense took place at University of Strasbourg in March, 2012.
