Comparative evolution is a powerful approach for understanding large biological data sets. During my PhD work, I used comparative evolution to identify RNA motifs within the core of the ribosome that are universally conserved or uniquely conserved in one phylogenetic domain of life (Bacteria, Archaea, or Eukarya). This work resulted in a provisional patent for novel drug targets. During my postdoctoral work at Harvard Medical School in Prof. Fred Winston’s lab, I investigated the information content of eukaryotic genomes, using yeast species as a model system. By integrating the most recent genomic methodologies, we discovered the existence of thousands of transcriptional promoters hidden within coding sequences (Doris, SM et al., Mol Cell, 2018). In optimal growth conditions these intragenic promoters remain dormant and thus have eluded prior detection. However, in the presence of genetic or environmental stresses, cells use these promoters to express alternative information, thereby expanding the coding potential of a single gene.
