• Designed and independently performed in vivo anti-tumor efficacy studies, ex vivo processing of lymphoid organs for analysis and adoptive transfer, and monitoring of tumor growth response and rechallenges in immunocompetent mice. Validated the generation of adaptive immunity with pharmaceutical test compounds.• Designed flow cytometry panels and performed flow analysis on murine tumors and tissues to characterize immune cell composition.• Implemented in life survival studies involving a luciferase-expressing lung tumor metastases model to monitor the anti-tumor efficacy of immune-modulating compounds.• Performed luminescence IVIS imaging of IFNb-reporter mice to complement nucleic acid-sensing projects.

• Determined anti-LAG3 to be superior to anti-PD-L1 as a supplemental antibody monotherapy of transgenic CD4 T cell, resulting in significantly fewer relapses in wild-type mice.• Identified a greater negative role of host PD-L1 in the exhaustion of tumor / self-antigen specific T cells than that induced by tumor-expressed PD-L1. Increased survival of tumor-bearing mice when made deficient for Treg cells and host PD-L1.• Created tumor / self-antigen, TRP-1, deficient wild-type mice to assess the effect of tumor antigen alone against shared tumor / self-antigen on CD4 T cells in immunocompetent mice.

• Obtained a DoD Post Doc Visionary Award grant that funded work published in the Journal of Immunology, cited greater than 100 times.• Identified exhaustion of transgenic tumor / self-antigen specific CD4 T cells with flow cytometry and cytokine / chemokine ELISAs.• Planned and conducted in vivo studies on the reversal of T cell exhaustion to treat relapsing melanoma with inhibitory receptor antibody therapy. Determined anti-PD-L1 and anti-LAG3 dual antibody therapy to be successful in mediating treatment of relapsing melanoma.• Developed and optimized a more reliable model of melanoma relapse, thereby greatly decreasing the number of mice needed to obtain consistent recurrence and amount of time for relapse to occur - recurrence rate went from 30-50% in weeks to months to 80-100% between 20-30 days.
