About Stephen Ledet

Originally from the Washington D.C. area, I am a rising junior at the University of Chicago and am pursuing an undergraduate degree in the Pritzker School of Molecular Engineering on the bioengineering track. My interests in academia lie heavily within the cross-disciplinary nature of the sciences and engineering. I am especially fascinated by the latest research on CRISPR-Cas9 and its gene editing implications.Biomedical research is another passion of mine. I collaborated with a professor of Global Health on research, of which the subject has demonstrated to me the much-needed cooperation between medicine and the humanities to reinvent healthcare in the future. Within the University of Chicago’s Prototype for Success program, I highly anticipate personalized workshops on the intersection of STEM and entrepreneurship from expert mentors and funded research experiences to refurbish my ability to think and act as a problem-solver and innovator.

The Chevrier Lab is an integrative immunology research team whose goals ultimately aim to understand the complexities of the organism’s immune response to combat infection or disease. This pursuit will hopefully uncover pieces to the puzzle in manipulating the immune system at will, revolutionizing the world of medicine and biomedical research.I spent 8 weeks with the Chevrier Lab through the Pritzker Scholar Match Program. My position on the team as an undergraduate research assistant was to catch up on the experiments my mentor and PhD student had been running before branching off on my own related project. The goals of my project fit within the tRNA “group” of the team— my experiments were specifically examining the relationship between the innate immune response and post-transcriptional modifications of certain tRNA species. My mentor also was exploring the combinatorial effects of adjuvants— I stimulated my innate immune cells with one of the single adjuvant treatments extensively studied.I performed a number of protocols and tasks in the duration of my internship. These included:- gRNA Cloning- Miniprep Purification- Bone Marrow Dendritic Cell (BMDC) Extraction from Cas9 Mice- Lentivirus Production in 293T Cells- T-Cell Extraction from OTII+ Mice- Coculture of BMDCs w/ T-Cells- Surveyor Nuclease Assay- Polysome Collection- Flow Cytometry Analysis (to assess T-Cell Proliferation)
