Discovery Pharmaceutical Sciences (DPS) is a multi-disciplinary research group that enables drug discovery and drug product development through application of pharmaceutical science and drug delivery research from target identification through clinical candidate selection. As a Senior Scientist in Merck's DPS-Boston team, I'm responsible to contribute into various formulation development efforts for diverse modality of pharmaceutical products.

✔ Expertise in Microfluidic preparation of Lipid Nanoparticle (LNP) formulations (using Precision NanoAssemblr Ignite) followed by Purification techniques and routine Analytical Characterization of Nucleic acid-encapsulated LNP formulations using DLS, Ribogreen, Quant-iT.✔ Supported Extended Characterization of LNPs using NanoFCM, AF4, Cryo-EM, NMR & assays like TNS, Release Kinetics.✔ Proficient in In vitro screening to identify 'Lead' candidates using various analytical assays (LDH, QUANTI-Luc), along with Flow Cytometry, PCR & Fluorescence imaging.✔ Performed Formulation Stability analysis in different storage conditions, Designed & executed thorough screening of cryoprotectants for Frozen Formulation (along with lyophilization) to ensure Long-term Storage.✔ Provided end to end support in formulating & optimizing LNPs under sterile condition for 30+ Preclinical In vivo studies in Mice & Non-Human Primates (NHP) for multiple program needs to deliver into various organs and delicately maintained Electronic Lab Notebook (ELN), Batch Manufacturing Record (BMR), Certificate of Testing (CoT).✔ Played a pivotal role in setting up SalioGen’s in-house Polymer Synthesis capability and assisted to set up several new instruments (SOPs) in 'Novel Drug Delivery' (NDD) team.✔ Conjugated small molecules, peptides, etc. on optimized nanoparticle for enhanced Targeted Delivery.✔ Collaborated with cross-functional teams to progress multiple projects with time-sensitive deliverables.✔ Contributed significantly in various discussions with collaborators i.e. CROs by providing guidance in synthesis & purification of novel molecules, conjugates.✔ Analyzed complex dataset using GraphPad Prism and communicated through scientific presentations in various levels of weekly, or monthly meetings.✔ Managed a thorough chemical inventory, conducted monthly audits to assure laboratory safety as part of SalioGen’s 'Chemical Safety' team.✔ Part of SalioGen’s 'Culture & Core Value' team.

1. Developed a novel cost-effective, biocompatible, hydrogel nanosensor for detecting ionizing radiation, used in clinical radiotherapy for cancer treatment and closely worked in collaboration with medical professionals at Banner M.D. Anderson Cancer Center & Arizona Veterinary Oncology in Gilbert, AZ.★ Our research on radiation dosimetry has been highlighted and featured in EurekAlert.2. Designed & optimized a library of aminoglycoside-derived lipid-based Polymeric nanoparticles • Characterized formulations for purity, long-term storage stability, using hydrodynamic diameter, zeta potential, and studied drug encapsulation/ release using analytical methods. • Screened formulations for nucleic acid & drug delivery using in vitro cell-based assays. • Initiated collaboration with Massachusetts General Hospital for testing their hepatotoxicity. 3. Formulated biomaterial-based liquid ink for clinical applications in endoscopic imaging • Characterized ink formulations by hydrodynamic diameter, PDI, zeta potential and tested its ex-vivo efficacy of injectability, contrast, spot-size using porcine intestinal tissue. • Utilized in vitro macrophage-based assay and in vivo mice model to evaluate inflammation & potential toxicity of synthesized ink-formulations. • Initiated collaboration with Mayo Clinic for in vivo pre-clinical trials of endoscopic imaging in pig.★ Selected for a press release during the ACS Spring-2022 meeting.4. Synthesized library of amino acid, lipid and small molecule drug-conjugated microbead for preferential binding of methylated and unmethylated DNA with the applications into bioprocessing & disease detection and collaborated for computational QSAR study with Rensselaer Polytechnic Institute.5. Synthesized & modified polymer biomaterials (e.g. silk, chitosan, gelatin) to make hydrogel-based adhesives/scaffolds, tested by collaborators for ex vivo & in vivo evaluation of diverse tissue sealing and wound healing applications.
