Kinesin Tail Domain (KTD): A Potential Therapeutic Candidate Against Hypertriglyceridemia | Thesis Supervisor: Prof. Roop MallikMy work explores the potential of targeting very low-density lipoproteins (VLDL) to effectively manage hypertriglyceridemia. I discovered that Kinesin-1, a motor protein, plays a crucial role in VLDL secretion by binding to triglyceride-rich lipid droplets (LDs) in the liver. Through all-atom molecular dynamics (MD) simulations, in vitro experiments, and cell culture assays, I demonstrated that the Kinesin-1 tail domain (KTD) preferentially binds to monolayer LD membranes. This binding disrupts LD distribution in hepatocytes and significantly reduces secreted TG levels without affecting other cellular functions. These findings highlight the potential of KTD as a therapeutic target to modulate lipid droplet dynamics and lower serum triglyceride levels, offering a promising alternative to current treatments. This research underscores the importance of innovative approaches in addressing complex metabolic disorders and advancing healthcare solutions.
