I have completed my doctoral studies under the supervision of Prof. Javed N Agrewala on the topic “Boosting the efficacy Bacillus Calmette–Guérin (BCG) vaccine against Mycobacterium tuberculosis” from CSIR-Institute of Microbial Technology, Chandigarh, India. During my doctoral training, I have tried to generate a vaccine to enhance the protective efficacy of BCG. We have generated a DNA vaccine comprise of 6 mycobacterial immunodominant T cell epitope peptides to enhance the Mtb specific T cell response. We have demonstrated that the combination of DNA vaccine and BCG decreased the Mtb burden in lungs as well as spleen, compared to BCG alone. The protection was provided by enhanced CD4 and CD8 T cell memory evidenced in both lungs and spleen. The enhancement in IFN-γ and decreased IL-10 were also observed through ELISA, indicating Th1 immune response. Further we have demonstrated the increased activation efficacy of antigen presenting macrophage and dendritic cells. Such approach can be used to improve the potency of BCG in the individuals. I have expertise in wide range of fields (immunology, infection biology, stem cell research, molecular biology, protein science and bioinformatics), working with primary cells (macrophages, Dendritic cells, B cells, and T lymphocytes, Fibroblasts), cell culture with mouse and human cell lines, animal models (Mtb infected Murine models), ELISA, Western blot, flow cytometry, MACS cell sorting, confocal microscopy, RT-qPCR, electrophoretic mobility shift assay, protein purification, transfection and transformation, immunohistochemistry, immunoinformatics and in silico tools used to check sequence as well as structural mimicry and association with autoimmune diseases. Because of the ability to work in a group, friendly nature and understandings, I was involved in different projects in the lab.
