Research ScientistUniversity Of Virginia Mar 2015 - Mar 2017Charlottesville, Va, UsObesity, Diabetes and Cancer Research.Following were the projects I worked in the Department of Pharmacology in the University of Virginia: 1) Determining the molecular regulation of fatty acid metabolism in the liver, 2) Determining how diet and obesity drive liver cancer initiation and progression, 3) Determining whether the metabolic pathway of lipogenesis can be therapeutically targeted to prevent or treat alcohol-induced liver cancer, and 4) Identification of small molecule drugs that increase mitochondrial metabolism for the treatment of obesity and metabolic disorders. I used a wide range of in-vivo and in-vitro models in these projects. These included knock-out mice models with tissue specific genetic knockouts, assays using primary hepatocytes, cancer and non-cancer cell lines and metabolic and immunoassays using serum and plasma. The techniques involved were metabolic assays using various radionucleotides, cellular oxidative capacity using SeaHorse XF analyzers, ELISA, Immunoblots, Immunohistochemistry, Real-time PCR, 2D and 3D cell cultures. -

For the first time performed detailed biochemical characterization of all six mammalian ceramide synthase. Discovered FTY720 to be a novel inhibitor of ceramide synthase and characterized the mode of inhibitory action. Developed biochemical assays for analyzing neutral lipid composition of mouse liver. -
