Bacterial Artificial Chromosome (BAC) libraries were used for human-genome sequencing and for the creation of knockout mouse models.My job functions included the following.• Library Screening. Performed hybridization using radio labeled synthetic Oligonucleotide probes.• Quality-Assurance of Knockout Mouse Models. Performed Gateway reactions, PCR and prepared Glycerol stocks for further research.• Standardize Fosmid Library Pool Strategies. Standardization of pool strategies to construct Pine Fosmid libraries. Performed serial dilution of the harvested cells at different concentrations to determine optimal pool size.• Picking Individual clones. Responsible for accurately streaking and picking specific clones from various libraries as LB STAB cultures (200 clones/day).• Characterization of BAC-clones. Performed restriction enzyme digestion and Pulse Field Gel Electrophoresis (PFGE) of digested BAC DNA to study average DNA insert size. • BAC Library Production. Responsible for colony picking of complete genomic libraries using Q-Pix Robots into 384-well format micro titer plates.• Re-arraying BAC-clones from 384 to 96-well format. Programmed and operated Q-Select robots to pick clones (600 clones/day), transferred into 96-well format micro titer plates.• BAC-clone Replication. Programmed and operated Q-Select robot to replicate BAC and Fosmid libraries into 384 well plates.• Macro array (Gridding). Generated macro arrays of BAC and Fosmid libraries on high-density membrane filters using BioGrid robots. The macro arrays were used for library screening, gene expression and gene discovery.
