Postdoctoral Fellow- Cardiomyopathy Research ScientistStanford University Oct 2011 - May 2016Stanford, Ca, UsProgram: Cardiomyopathy and Heart Failure• Project leader in developing assays to understanding how the tunability of the cardiac contractility is affected by the interaction of the myosin enzymatic motor domain with both myosin-binding protein C (MyBPC) and myosin S2 domain in both wild type and cardiomyopathy-causing myosin mutants. • Supervised a project which involved characterization of the enzymatic properties of different cardiomyopathy-causing cardiac myosin mutants in understanding their role in human cardiomyopathies. Performed bulk enzyme kinetic assays and in vitro motility experiments to measure the rates of the various steps of the myosin ATPase cycle. Used single molecule force spectroscopy to characterize the force generated by a single myosin molecule. Finally using loaded in vitro motility showed that the power output of individual myosin molecule is altered because of these mutations. • Directed the efforts of understanding the effects of phosphorylation on the biomechanics of human cardiac myosin. Using force spectroscopy showed that the internal power producing ability of a single myosin head is enhanced upon the phosphorylation of the regulatory light chain associated with it. • Drove a project of understanding the cardiomyopathic causing mutations in the troponin T complex of the regulated thin filament. Used the bacterial expression system to purify the troponin complex proteins. Used steady state fluorescence and stopped-flow kinetic measurements to measure the dynamics of Ca2+ binding and showed that the Ca2+ dissociation kinetics is altered in these mutants. -
