PrimeraDx is a molecular diagnostic manufacturer with full service companion diagnostic (CDx) product development capabilities. I was mainly involved in• Developing and verifying tests for oncology studies on an automated PCR-capillary electrophoresis platform.• The project involved single-tube 18-target, multimodal quantitative Copy Number Variation and gene expression panel that can detect: Gene copy number variation (CNV), Chromosomal copy number variation and Over-expression of mRNA.• Designing and developing positive controls for PCR and RT.• Sample preparation optimization • Performing validation and verification studies• I was also extensively involved in transferring the assay to manufacturing team for a RUO product, which includes: generating quality control procedures, performing incoming QC for reagents, manufacturing reagents and transferring a 18 target quantitative RT-PCR assay.Publication/Posters:Jork Nolling, Kiran Madanahally Divakar, Supriya Shekar, Amy Leblang, Jessica Riley, Lilly Kong. I was involved in developing molecular diagnostic detection tests for NIH awarded Grants for infectious diseases.• Designed multiplex primers and probes for Immunetics’s Reverse-line blot technology for several infectious disease projects. The projects includes: Bacterial identification and antibiotic resistance panel, Trypanosoma and leishmania panel and Fungal diagnosis• Cultured and maintained several antibiotic resistance bacteria for verification and validation studies.• PCR optimization, hybridization and detection using Immunetics’s Reverse line blot technology • Developed resistance panel, which includes: Ampicillin resistance, Carbapenemase (includes recent super-bug NDM), ESBL, MRSA, VRE, Fluoroquinolone and Aminoglycosides resistance.• Also developed LAMP based assay for fungal detectionPublication/Posters:Supriya M.
