The second objective was to compare these formulations in terms ofLoading of quercetin, toxicity to HaCaT (keratinocytes) THP-1 cells (Monocytes) and Vero (epithelial), and finally the maintenance of the activity of quercetin on cells in vitro, in order to demonstrate an increase in skin penetration of Quercetin in vivo.In this project, three approaches to nanometric formulations (smartCrystals®, Lipid nanocapsules and liposomes) were tested to improve the solubility of quercetin. The formulations are optimized in terms of preparation process (Industrial transposition) and of composition of the excipients to increase the quantity of formulated quercetin.The formulations were characterized on several parameters: size, PDI, loading In quercetin, crystalline state and kinetics of quercetin release in vitro. Then, theFormulations were compared with each other on HaCaT and THP-1 cells with determinationof their toxicity and protective activity. Finally, two formulations (quercetin smartCrystals® withTPGS and quercetin LNC 20) were selected and compared in vivo to evaluate The improvement of skin penetration of quercetin. -
