TEACHING AREAS OF INTEREST/SPECIALIZATIONMetabolic System Biology (Metabolomics and fluxomics), Fundamental of Biochemistry, Bioseparation technology, Advance analytical techniques, Intermediary Metabolism and regulation, Secondary metabolitesRESEARCH INTERESTSMetabolomics, Metabolite profiling using stable isotope, 13C-Metabolic flux analysis (13C-MFA), Secondary metabolites, Volatile organic compounds RESEARCH EXPERTISE• General: Biochemistry and Molecular Biology of primary and Secondary Metabolism• Specific: (1) Metabolic Profiling in biological system (plants and microbes)(2) Mapping cellular metabolic pathways in microbial system using 13C stable isotopes(3) 13C-Metabolic Flux Analysis (13C-MFA) through GC-MS analysis technique.(4) Reconstruction of metabolic pathway of microbial systems using genome-based annotation.(5) Extraction of metabolites (primary and secondary) from biological samples (tea, rice grain) for the analysis (qualitative and quantitative) by GC-MS and HPLC(6) Derivatization of metabolites (sugars, amino acids, organic acids and fatty acids) for GC-MS analysis, followed by analysis and identification of acquiring data through software like MassHunter (Agilent Technologies) and GCMS solution (Shimadzu) equipped with NIST Mass Library.(7) Isolation and identification of soluble and cell wall bound phenolics from plant(8) Extraction or trapping of volatile compounds through static headspace (SPME) and dynamic headspace techniques from microbial and plant samples (in-situ and ex-situ condition).(9) Enzyme purification (Gel filtration chromatography, Ion exchange chromatography, SDS-PAGE, Native Gel)(10) Enzyme assay and kinetics (Km, Vmax, Kcat)(11) DNA, RNA and plasmid DNA isolation from biological system (Plant and microbes)(12) Synthesis of cDNA from isolated mRNA and cloning of core cDNA into vector(13) Design of degenerate primer (Sequence homology based) and gene specific primers(14) Cloning of plant genes and semiquantitative transcript expression analysis through PCR(15) Extraction, purification and characterization of oligosaccharides from grain crops(16) Isolation and characterization of microbes (including microbial techniques)(17) Method development, standardization and validation• Practical Expertise: HPLC (Waters Corporation), GC-MS (Agilent Technologies and Shimadzu), FTIR (Shimadzu), semiqRT-PCR (Mastercycler, Eppendorf) Gel electrophoresis (Bio-Rad Laboratories)• Statistical analysis: PCA (by MetaboAnalyst software), ANOVA and DMRT (by SPSS software)
