Cancer is generally described as a multistep process characterized by uncontrolled malignant growth of cells, leading eventually to metastases that account for most of the human fatality cases. Hence, understanding the mechanisms that promote tumorigenesis and metastasis is an important aspect for developing therapeutic strategies against cancer. In previous years, our lab has generated a simple in vivo model system using Drosophila melanogaster that we use to investigate genes which cooperate with known oncogenes and thereby lead to epithelial transformation and metastasis.In my PhD project, I aim to identify genes and cellular processes with previously unidentified roles in tumorigenesis and cancer progression. In particular, I am investigating genes cooperating with known oncogenes pathways, i.e. EGFR signaling, in tumor formation using a variety of approaches such as confocal imaging, FACS, invasion assays and expression profiling. This work will expand our knowledge of fundamental genes and cellular events contributing to cancer and will therefore have broad implications for understanding human development and disease.

Tissue homeostasis requires a tightly regulated balance between cell death and proliferation, which is often surveyed by somatic stem cells residing within the tissue. A critical mediator of key signaling events during adult homeostasis and development is the highly conserved Wnt family of secreted proteins that activate different downstream signaling cascades. Aberrant regulation of Wnt signaling is a prevalent theme in numerous diseases such as cancer. Although recent discoveries of canonical Wnt signaling components as well as insights into Wnt secretion and trafficking have increased our understanding of how Wnts signal, the complex mechanisms of many essential events during Wnt signal transduction still need to be solved. In this study, we identified a new regulator of the Wnt signalling pathway and characterised its function in vitro and in vivo.
