Single cells multiplex proteomic analysis with cleavable fluorescent tyramide (CFT), Supervised by Dr. Jia GuoLocation: Arizona State University, Tempe AZ-- This project aims to generate cell type distribution map of Tonsil tissue by the detection and analysis of 20 different proteins in every single cell using cleavable fluorescent tyramide. -- Synthesized the cleavable fluorescent tyramide featuring the azide based cleavable linker.-- Develop new cleavage condition that remove 90-95% of fluorescent signal. -- Collect imaging data, process data analysis, and visualize data using viSNE dimensions reduction algorithm.

Highly multiplexed single-cell in situ protein analysis using cleavable fluorescent antibodies -- Coupled an azide-based linker to fluorophores and used it to label antibodies-- Developed a highly multiplexed single-cell in situ protein analysis method though cyclic immunofluorescence with cleavable fluorescent antibodies-- Achieved multiple protein staining in single cells-- Analyzed high throughput, high dimensional protein expression cell data, studied single cell heterogeneity and applied hierarchical clustering to study signaling pathways
