During my time as the Director the University of Michigan Transgenic Animal Model Core my group has established and optimized protocols for the production of genetically modified mice and rats for biomedical research. We routinely use genome editing nucleases (CRISPR/Cas9, zinc finger nucleases and TALE nucleases) to produce gene knockout and gene knockin models. The Core offers a CRISPR/Cas9 pipeline in which the investigator names the desired genetic change and Core personnel take all steps necessary to design, test, and produce the desired mouse or rat gene modification with CRISPR/Cas9 technology. The Transgenic Core guarantees the production of transgenic mice and rats by random integration of transgene DNA molecules. The Transgenic Core guarantees mice and rats with gene knockouts with reagents designed and tested in the Core. We have generated over 18,000 transgenic mouse founders and more than 1000 transgenic rat founders. In addition we offer bacterial artificial chromosome recombineering and gene targeting in mouse ES cells. We provide a complete suite of assisted reproductive technologies and cryopreservation services.

This includes 1) the production of transgenic mice, 2) the production of embryonic stem (ES) cell-mouse chimeras, 3) production of transgenic mice, 4) production of transgenic rats, 5) preparation of genetically modified mice and rats through the use of zinc fingers, 6) de novo preparation of mouse ES cell lines, 7) hands-on training in all technologies, 8) support services, 9) and consultation on all phases of transgenic and gene targeting research from experimental design to mouse breeding.Since 1989 the Transgenic Core has produced over 15,000 transgenic founder mice from more than 1,500 transgenes and nearly 1,000 transgenic rats. Gene-targeted (knockout) mouse strains have been successfully generated from 226 gene targeting vectors in collaboration with other investigators. 200 ES cell clones have been imported from the International Mouse Knockout Consortium and numerous mouse lines generated from them.
