(Hons) in Biomedical Science s in 2014 and then completed my Master's in Science (M.Sc.) at the University college of London (UCL) in Biomaterials and Tissue Engineering in 2016. At UCL, I used my thesis to prove that ultrasound has the potential to revolutionize 3d printing. After completing his M.Sc., Thomas worked in cutting edge life sciences manufacturing, most notably as the production lead at the ex-Wolfson Gene Therapy Unit, which eventually became part of MeiraGTx. While in Malta, he has spent the last 5 years employed as a Medical Scientific Liaison educating local health practitioners on how, when and why to use medical cannabis. Immune checkpoint blockade relies on a pre-existing inflamed tumor microenvironment and a pre-existing immune response to tumor neoantigens for clinical activity, which are only present in some patients. Oncolytic immunotherapy can provide both of these and therefore combination of these two proven modalities is particularly logical and attractive.Replimune is developing novel, proprietary oncolytic immunotherapies intended to improve both the direct anti-tumor effects of selective virus replication and the potency of the immune response to the tumor antigens released. As part of the production team, I was co-responsible for both upstream and downstream processes essential for the production of gene therapy products.A significant aspect of my role included actively participating in the optimization of the unit's Standard Operating Procedures (SOPs) to achieve full compliance with GMP standards. This entailed raising Quality Incident Reports to initiate change controls for the amendment of non-compliant SOPs. Additionally, I contributed to SOP re-validation efforts to ensure repeatability and consistency in production processes.My involvement in the production process was comprehensive, encompassing various laboratory-based tasks. This included cell culture, transfection with plasmids, treatment with benzonase, cell rupture using microfluidization, AKTA chromatography purification for virus elution, concentration and diafiltration using a TFF module, and sterile vialing of the product in preparation for clinical trials.Through my hands-on involvement from start to finish in the production of gene therapy products, I developed invaluable skills and expertise in GMP-compliant manufacturing processes.
