Carried out a research project exploring the mechanisms responsible for low ethanol yield in wheat. The properties of protein, starch and starch granules in the wheat grain were studied extensively using a variety of techniques. These included; SDS-PAGE, peptide fragment fingerprinting, enzymatic assays, mass spectrometry (ESI/MALDI), microscopy and x-ray diffraction. Using these techniques it was possible to conclude that the starch granule structure and starch molecular structure of the two wheat varieties was different. The identified phenotype would allow wheat varieties to be screened for their ability to produce glucose from starch.
