My early cell biological work critically contributed to the notion that locally coalesced membrane raft domains function as platforms for membrane-proximal cell activities, like surface receptor signaling or cytoskeleton-membrane interactions. Based on this concept research teams which I led at the Basel Institute for Immunology and at the Sir William Dunn School of Pathology at the University of Oxford, characterized cell biology, biochemistry, biophysics, and immunology of plasma membrane rafts which control activities of T cells.Consequent translation of this concept toward medical application defined T-cell plasma membrane rafts as target for a regulatory signal constellation which induces a dormant state in resting T cells under which all T cell responses to activating TCR stimulus are comprehensively suppressed.
