Coli, following genotoxic stressMethods : Construction of mutants, cloning, plasmid DNA extraction, PCR, sequencing, microscopyFaithful duplication of the genome is necessary to ensure genomic integrity throughout generations. However, many DNA damages occur sometimes spontaneously or because of a genotoxic stress, and this represents a serious threat to genome stability. DNA double-strand breaks, induced by drugs like mitomycine C, are serious genomic lesions that are potentially lethal at the cellular level. A repair mechanism of the lesion proceeds through homologous repair, which is based on sequence homology, DNA compaction and induction of the SOS response.The aim of the internship was to create various mutants in Escherichia Coli with deleted genes involved in repair mechanism : one of these is called sbmC, encoding for a gyrase inhibitor. Thanks to a genetic tool custom-made by O. Espéli's team, it was possible to reveal the effect of a gene deletion on sister chromatid cohesion and understand more precisely the role of these genes. Moreover, I analyzed one of the main essential recombination proteins RecN. By fusing the gene with a GFP cassette, I optimized a protocol to locate it in fluorescence microscopy.
