Goal: talk about science in a relaxed atmosphere conducive to sharing and discussion.Main tasks and skills acquired in self-study :• Produce quality live broadcasts and master technical details: managing sound, microphone, lights.• Prepare, schedule and conduct interviews with guest scientists.• Master video encoding (OBS Studio), editing (Adobe Premiere Pro), graphic design (Canva) and social media management (Hootsuite) software.• Host live chats (approximately 2h-2h30) by interacting with the chat.• Feed all social networks to promote the shows (Facebook, Instagram, Twitter).• Manage the accounting related to this activity to purchase equipment and improve the quality of the shows.• Creation of 26-hour non-stop shows in May 2021 & May 2022 with a series of scientific interviews and discussions. M.Sc. student in Dr Espéli's lab - Chromosome dynamicsProject title : Study of DNA repair in E. Coli, following genotoxic stressMethods : Construction of mutants, cloning, plasmid DNA extraction, PCR, sequencing, microscopyFaithful duplication of the genome is necessary to ensure genomic integrity throughout generations. However, many DNA damages occur sometimes spontaneously or because of a genotoxic stress, and this represents a serious threat to genome stability. DNA double-strand breaks, induced by drugs like mitomycine C, are serious genomic lesions that are potentially lethal at the cellular level. A repair mechanism of the lesion proceeds through homologous repair, which is based on sequence homology, DNA compaction and induction of the SOS response.The aim of the internship was to create various mutants in Escherichia Coli with deleted genes involved in repair mechanism : one of these is called sbmC, encoding for a gyrase inhibitor. Thanks to a genetic tool custom-made by O. Espéli's team, it was possible to reveal the effect of a gene deletion on sister chromatid cohesion and understand more precisely the role of these genes. Moreover, I analyzed one of the main essential recombination proteins RecN. By fusing the gene with a GFP cassette, I optimized a protocol to locate it in fluorescence microscopy.
