I am driven to help patients through research. Over 10 years of work in oncology and immunology, I have developed numerous diverse approaches to cancer therapy, including the examples below and further undisclosed projects.• Used epigenetic modulating agents to upregulate cancer-testis antigens (CTAs) and antigen processing machinery, increasing efficacy of CTA-based vaccines. • A small molecule LSD1 inhibitor targeted hormone/growth signaling as well as cells of the TME, including pro-inflammatory M1 macrophage polarization. A PROTAC variant led to targeted protein degradation.• Depleted anti-inflammatory cancer-associated fibroblasts with prodrugs activated by the protease activity of fibroblast activation protein.• Used IV-infused mesenchymal stem cells to deliver internalized cytotoxic agents.• Inhibited anti-inflammatory IL-4/IL-13 signaling on macrophages.• Inhibited pro-tumorigenic miRNAs.I also have 2 years of experience in other immunological disorders such as EAE (MS), lupus, arthritis, and allergy. I have done immunophenotyping and functional assays with innate and adaptive immune cells including NK cells, T cells, Tregs, B cells, plasmablasts, monocytes/macrophages, conventional and plasmacytoid DCs, neutrophils, and fibrocytes. I am proficient with mouse tissues (syngeneic/xenograft models, primary cells, cell lines) and human tissues (PBMCs, whole blood, cell lines).Over the course of such projects, I gained extensive drug development experience in areas such as target validation, in vitro assay development, in vivo xenograft/syngeneic mouse models, biomarker discovery, and pre-clinical PK/PD with a proven track record of success. Over 7 years of undergraduate/graduate research, I earned authorship on 14 publications and drove a project to an upcoming clinical trial. In the industrial setting, I have completed assay development for 4 projects entering clinical trials, including report writing for IND submission.Conventional and spectral cytometry is my greatest technical strength, featuring 8 years of experience across 5 instruments. I have performed numerous mouse immunophenotyping studies, including dissection of tissues (tumor, spleen, lymph nodes, bone marrow, thymus) and training of 7+ people across 5 different laboratories. At CellCarta, I led an 8-person team to develop deterministic, data-driven approaches for effective panel design and used these approaches to create novel large (up to 35c) modular panels, a pivotal part of the company’s push into spectral flow cytometry services.

    • Spearheaded a highly collaborative project between our academic lab and the CEO and scientists of Imago Biosciences to develop and characterize a novel inhibitor of an epigenetic enzyme, LSD1.• Tested the hypothesis that pharmacologic and genetic inhibition of LSD1 can be effective against lethal castration-resistant prostate cancer independent of androgen receptor signaling by using 10+ syngeneic or xenograft mouse models, including patient-derived xenografts and genetically-modified (CRISPR, shRNA) cell lines.• Used ex vivo and 3D culturing experiments to demonstrate direct effects of LSD1 inhibition on cells of the tumor microenvironment, including inhibition of endothelial sprouting and promotion of M1 macrophage polarization.• Designed and optimized six 8-12 color flow cytometry antibody panels used to identify lymphocyte, myeloid, and stromal populations in large mouse experiments. Trained 5+ people to perform tissue collection (tumor, spleen, lymph nodes, bone marrow, thymus), antibody staining, and data analysis. 4-5 collaborative publications are expected resulting from this work.• Developed a protocol for isolating and expanding murine bone marrow-derived mesenchymal stem cells (MSCs).• Utilized MSCs as delivery vehicles for cytotoxic agents to tumors. Longitudinally tracked homing of IV-infused, 89Zr-labeled human MSCs in mice using µPET imaging.• Performed and trained others to perform ex vivo human T-cell co-culture experiments to functionally analyze T-cell growth and activation.• Chosen as the 2019 Johns Hopkins applicant for the PhRMA Foundation Predoctoral Fellowship, and was then selected among national applicants to receive a two-year, $40,000 fellowship.• Demonstrated desire for knowledge and self-improvement by taking one elective course each semester of the final three years during my thesis work.• Gained extensive cancer cell line culture experience, including viability, clonogenic, and cell cycle assays.
