Phd Candidate
This AstraZeneca-funded Ph.D. project in the Department of Renal Medicine at University College London pursued a translational approach to address mitochondrial dysfunction in kidney disease and was supervised by Prof. Robert Unwin (UCL/AstraZeneca) and Prof. Jill Norman (UCL). This exciting and versatile project allowed me to develop a unique technical and soft skill set. I performed extensive mechanistic studies in cell lines and primary cells using techniques such as molecular biology techniques (qPCR, WB), immunostaining (IHC/IF/ICC), cell-based assays (viability, toxicity, bioenergetic profiling (Seahorse Analyzer)), gene editing and silencing (CRISPR/Cas9 and RNAi), confocal super resolution imaging and transmission electron microscopy. I also carried out two in vivo studies of acute kidney disease (ischaemia/reperfusion injury) and chronic kidney disease (folic acid nephropathy).Key achievements:- Successful grant application - Initiation of multiple collaborations with KOLs - Supervision of undergraduate research projects- Ad hoc reviewer for high-impact journal- GCP certified
Graduate Research Student: Master Thesis
This Master thesis project was carried out in the Max-Rubner laboratory of the German Institute of Human Nutrition (DIfE) in collaboration with the Experimental and Clinical Research Center (ECRC) of the Charite. The main aim of this project was to investigate the role of the ion channel TRPC6 in obstructive nephropathy and was supervised by PD Stephanie Kraemer (DIfE) and Prof. Maik Gollasch (ECRC, Charite). I carried out unilateral ureteral obstruction (UUO) surgeries in WT and TRPC6ko mice to investigate the impact of TRPC6 ablation on disease progression. Subsequently, I performed histological stains (PAS, Sirius Red), immunodetection methods (IHC/IF) and performed molecular biological techniques (qPCR/WB) to detect tubulointerstitial damage, immune infiltration and fibrosis. Furthermore, I isolated glomeruli from UUO and sham kidneys and characterized the effects of TRPC6 ablation on glomerular injury using qPCR and WB.Key achievements:- published a review article - published a primary research article

During this graduate research project in the group of Prof. Schuelke-Gerstenfeld (Department of Neuropediatrics, Charite), I developed a laser capture microdissection (LCM, Arcturus XT) protocol for the dissection/ isolation of revertant fibres from muscular dystrophy (MD) patient-derived biopsies as well as a sequencing protocol. Furthermore, I characterized MD patients by IHC/IF to investigate the underlying processes involved in reversion and the role of dystrophin in satellite stem cell biology.Key achievements:- developed a laser-capture micro dissection protocol for isolation of revertant fibers from patients with gamma sarcoglycanopathy. - developed a sequencing protocol to identify mutations in SGCG gene
Graduate Research Student
During the research internship in the group of Prof. Regitz-Zagrosek (Center for Cardiovascular Research (CCR)), I developed a protocol for cardiac fibroblast extraction and culture from murine and rat hearts. Furthermore, I isolated, differentiated and characterized murine peritoneal macrophages and investigated the impact of estrogen on this process. I also investigated how estrogen stimulation influences fibroblast to myofibroblast transdifferentiation - in a sex-dependent manner. As last part of this project I worked analysed murine cardiac tissue from a CVB3-induced myocarditis model and applied histological stains and immunostaining to analyse sex-dependent mechanisms in dilated cardiomyopathy.
Trainee Research Development
In 2013, I worked as research trainee in the CardioMetabolic Diseases Research unit at Boehringer Ingelheim (USA) in collaboration with Yale medical school in 2013. This project which was supervised by Dr. Noelynn Oliver and focused on the role of an ion channel in Chronic Kidney Disease (CKD). This incredible opportunity allowed me to develop a large technical and soft skill set. Techniques I put to use included:- Complex in vitro culture systems- Tissue/cell culture especially assay development (protein trafficking, cell migration,...)- Tissue harvest/dissection, embedding and sectioning - Laser capture microdissection/ nucleic acid isolation and purification- Immunohistochemistry as well as immunocytochemistry - Viral transduction (BacMam) & Transfection - Fluorescence microscopy and Scanning (ScanScope FL/ ScanScope AT Turbo)Key achievements:- developed a laser capture micro dissection protocol for isolation of glomeruli from kidney samples- developed/optimized a viral transduction protocol for human podocytes
Undergraduate Research Student: Bachelor Thesis
This Bachelor Thesis project with the title "The investigation of active DNA demethylation processes during retinal maturation" was carried out in the Department of Pharmacology for Natural Sciences and supervised by Prof. Stylianos Michalakis. The primary focus of this project was the function of the methylcytosine dioxygenase TET3 which transforms 5-methylcytosine to 5-hydroxymethylcytosine, a key step of DNA demethylation.Methods I put to use were: - RT-qPCR analysis indicated epigenetic relevant enzymes during retinal maturation - Molecular cloning of shRNA constructs to deplete retinae of various epigenetic key enzymes- rAAV transduction in retina culture as well as subretinal injection in mice
