Synthesis, labelling and pharmacological properties of new 5-HT3 receptor ligands The work presented in this thesis concerns the study of ligands of central serotonin (5-HT) receptors. It has two parts: 1) 5-HT3 receptors are ion channels and 5-HT3 antagonists have properties (antiemetic, antipsychotic. . . ) that make them potential drugs. Taking the basic structure of zacopride, we have synthesized the first iodine radioligand of the central 5-HT3 receptors. 125I-iodozacopride has proven to be an excellent tool for autoradiographic studies of 5-HT3 receptors in the rat central nervous system. In addition, other zacopride derivatives were synthesized for photoaffinity labeling and for purification of these receptors from NG 108-15 cells.2) 5-HT1a receptors are the prototype of G-protein coupled serotonin receptors. Using molecular modeling software (MAD,TSAR), we attempted to correlate 5-HT1a pharmacological activity to a number of parameters derived from molecule structures (QSAR). For two homogeneous families (indoles and tetralines), a screen was established allowing to correctly classify eighty percent of the active or inactive molecules. Finally, this model has been successfully used to predict a possible recognition by 5-HT1a receptors for indoles and tetralines, but also for other chemical families.
