About Tom Cunningham

The overall focus of my research centers on understanding the mechanistic basis for the reprogramming of cellular metabolism upon activation of oncogenic pathways. Cancer cells display alterations in the uptake and utilization of nutrients that distinguish them from normal cells. The identification of these key differences may pinpoint vulnerabilities in cancer cells and provide a path for novel therapeutic intervention. The question then becomes, “How can we identify these key differences in the metabolism of normal cells versus cancer cells?” My research involves a multifaceted approach that utilizes the disciplines and tools of biochemistry, molecular biology, mouse genetics, metabolomics, and cell biology to elucidate the molecular underpinnings of disease etiology. I believe we can use the knowledge of signaling pathways and cellular processes known to be perturbed in cancer cells to discover distinct points of intersection with the metabolic network. For example, my PhD work utilized a computational genomics approach to identify a novel connection between the mTOR signaling pathway and the transcriptional machinery responsible for controlling the expression of mitochondrial genes important for cellular energy production. During my postdoctoral training, I found a functional link between the protein synthesis capacity of MYC-overexpressing cells and the nucleotide biosynthesis pathway. Importantly, loss of function of the key enzyme at the nexus of protein synthesis and nucleotide metabolism specifically kills cancer cells but not normal cells, therefore suggesting new therapeutics capable of targeting this key metabolic node may be powerful anti-cancer treatments. The ultimate goal of my research is to translate the basic science of the continued identification of these critical metabolic nodes into new drugs, diagnostic tools, and biological insights that can positively impact patient health and survival.
