The American Diabetes Association has found that 40% of diabetic patients will have kidney damage related to Type 1 diabetes. Transforming growth factor beta (TGF-beta) is a key cytokine whose signaling pathway causes many diseases, and can specifically cause disorders and damage to the kidney associated with diabetes in humans and rodents. When mutated or altered, the TGF- beta signaling pathway can cause a wide variety of human disorders. The Gaussia luciferase assay is our primary tool in assessing the SMAD protein-protein interactions along the TGF-beta signaling pathway. Our goal is to identify unique compounds targeted to SMAD proteins, a class of proteins that are phosphorylated and activated by the TGF-Beta receptors. Libraries of chemical compounds have been prepared for use with appropriate screening techniques to discover new drug leads. Some of these compounds may inhibit key proteins along the TGF-Beta signaling pathway and could provide much needed relief for kidney cells of diabetic patients. Our hope is that the discovery of specific chemical inhibitors of Smad protein-protein interactions may be developed into competent drug treatments.
