The aims of this study were to investigate the effects of (superoxide dismutase) SOD1 as well as Angiotensin-converting enzyme (ACE) in the white adipose tissue of PCOS, PCOS-EMPA, Control &… Show more I was accepted into the SURE (Student Undergraduate Research Experience) program at The University of Mississippi Medical Center to study under Dr. Licy Yanes Cardozo in the Cell and Molecular Biology Department. More specifically, I studied Polycystic Ovary Syndrome (PCOS) in female rat models. The aims of this study were to investigate the effects of (superoxide dismutase) SOD1 as well as Angiotensin-converting enzyme (ACE) in the white adipose tissue of PCOS, PCOS-EMPA, Control & Control-EMPA rat models.During my time in the lab, I performed western blot protocol in order to further characterize Polycystic Ovary Syndrome and why it is associated with high blood pressure. I learned how to use machinery such as an EchoMRI on 16-week-old Sprague Dawley (SD) rats in order to track fat mass, lean mass, free water, and total water mass. In addition, I effectively utilized standard laboratory equipment including hand pipettes, balances, scales, centrifuges, image lab 6, electrophoresis machine. I performed Bicinchoninic Acid Protein Assay in order to quantify the total protein of a sample, homogenized samples of kidney in order to later perform western blot and protein assay, and organized and prepared chemicals, as well as labeling tubes and cleaning dishes appropriately.In summary, I found that: 1. EMPA in PCOS causes profound increases in glucosuria.2. There was a decrease in fat mass accompanied by decreased leptin.3. No change in insulin sensitivity.4. Decreased medullar ACE expression and activity accompanied with mild decrease in MAP.5. EMPA could be a novel therapeutic for treating obesity and increased blood pressure in women with PCOS.
