Research Analyst

Mrc/Urvi

Uganda

I was honoured to work in a BSL3 laboratory under GMP conditions, as a research internunder the MRC/UVRI and LSHTM Research Unit in Uganda. The research project wasassociated with local hospitals where our samples were collected via post mortem study.Immunity against M.tb is localised at the site of infection, the lungs. Lung draining lymphnodes and bone marrow have also been shown to contain TB specific memory cells. TheTBPM study aimed to compare lung immune cells in active TB, against those in latent TBand those uninfected individuals using flow cytometry. However, analysis by flow cytometryis only possible for single cell suspensions, whereas cells in tissues only occur in an extracellular matrix. Therefore, these cells must first be released from the extra cellular matrix intoa single cell suspension. In order to isolate individual cells from the lung tissue, the tissueswere dissociated enzymatically and physically to liberate the cells from the extra cellular matrix. Enzymatic dissociation was achieved using a digestion mixture which containsCollagenase D and DNase I which digest extra cellular matrix and DNA respectively. Themixture was physically dissociated using a GentleMacs and passed through a sieve to obtaina single cell suspension. The suspension is purified to remove RBCs and non-mononuclearcells. PBMCs in the TBPM study were isolated from blood samples by density centrifugationusing lymphoprep as a cell separation medium for recovery of mononuclear cells. IsolatedPBMCs were used in subsequent analysis for cell surface staining, intracellular staining, etc.The remaining cells will be used for further studies.
