Cell Reports · Jul 12, 2022Thymic-resident macrophages, a rare population that only represents 0.1% of total cells in the thymus organ, play an essential role in removing apoptotic thymocytes and maintaining thymus homeostasis. However, it remains unclear how thymic-resident macrophages can efficiently engulf apoptotic thymocytes. Therefore, we sorted the primary macrophages and performed metabolomics, RNA sequencing, and single-cell RNA sequencing analyses. Together with the in vitro studies, our work demonstrates the thymic-resident macrophages overcome continuous efferocytosis-induced oxidative stress by up-regulating the PPP, which highlights the central role of the PPP in maintaining thymic-resident macrophage and efferocytotic macrophage functions.During the research, I formed the hypothesis of PPP supporting the neutralization of oxidative stress in the thymic macrophages. With Dr. Chia-Lin Hsu’s guidance, we designed the proof-of-concept experiments, including the breakthrough of the challenging ex vivo thymic explant treatment. Furthermore, to achieve the complex multi-omics analysis of the metabolism network, I had learned R programming to accomplish the overall understanding of thymic macrophage metabolism. Together with flow cytometry, primary cell sorting, immunofluorescence staining, mitochondria content measurement, and cell culture experiments, we have put all the pieces together to prove our hypothesis. Not only did I learn bioinformatics skills and perform experiments, but I had also built a complete study from scratch. After three years of hard work in the laboratory, we have set up a milestone in investigating the rare thymic population. Show less

After extensive discussions with paper-making experts, we have devised a plan to replace the chemicals used in traditional paper reprocessing and to increase recovery of paper fibers. In our model, the paper sheets undergo image analysis to determine which spots have ink printed on top and will be sprayed with Saccharomyces cerevisiae which has been engineered to express deinking enzymes upon light induction. Instead of treating all paper pulp, only the spots with ink is shone with light to induce localized deinking. This procedure can remove ink efficiently while protecting most of the paper fibers from being digested. This design extends the lifetime of the paper fibers and reduces wood-logging for making paper. Show less
