Individual project of investigation of leucine-rich repeat receptor kinase from Arabidopsis thaliana - PSY1R. PSY1R is a leucine-rich repeat receptor kinase (LRR-RK) from Arabidopsis thaliana, which binds PSY1, a sulphated secreted peptide. The receptor is shown to be involved in root elongation and of plants defence responses. Additionally it activates the proton pump AHA2 by phosphorylation. However the mechanism of activation of PSY1R is still to be found. Some putative phosphorylation sites were detected with mass spectrometry and activation loop gene alignment, but their function is still unknown. The aim of the project was to investigate the influence of these putative sites on the activity of the intracellular kinase domain of the receptor and its ability to get transphosphorylated.There were used techniques as site-directed mutagenesis, Gateway© cloning system, radioactive ATP assay, among others.
