Gibson III, Ph.D. at University of Florida College of Dentistry.• Abstract: Periodontal disease (PD) is a complex chronic oral inflammatory disease, which in severe cases leads to loss of dentition. The inflammatory response which characterizes PD is initiated by a dysbiotic subgingival plaque community. An array of bacteria have been detected in the subgingival plaque of individuals with PD. Among these bacteria, Porphyromonas gingivalis is recognized as an organism closely associated with PD. Macrophages (Mf) represent 5-30% of the immune cells that infiltrate chronic PD lesions; yet the precise function of these cells in the context of PD is poorly understood. Mf undergo classical (M1), or alternative (M2) activation to finely-tune the function of these cells at the micro-environmental level. We hypothesize there are differences in the immunologic responses of M1, and M2 Mf to P. gingivalis. The approach involved defining the cellular immune response of M1- and M2-activated Mf to P. gingivalis employing the human THP-1 cell line. Utilizing ELISA assays, we observed that M1 Mf expressed robust levels of tumor necrosis factor (TNF)-α and interleukin (IL)-6 levels to P. gingivalis. Interestingly, while M2 Mf expressed TNF-α similar to M1 Mf, these M2 Mf failed to produce IL-6 to P. gingivalis. Our data suggest that the inflammatory response of M2 Mf to P. gingivalis challenge is impacted by immunologic activation. -
