Ph.D. research, funded by Veridex LLC, a Johnson & Johnson company, in the Medical Cell BioPhysics group of Prof. Dr. Leon Terstappen. Development of an automated image cytometer, CellTracks TDI, which detects very rare circulating tumor cells, in varying stages, in the blood of cancer patients. The accurate detection and characterization of circulating tumor cells is crucial in the development of personalized therapies for individual cancer patients.The developed image cytometer uses a TDI camera to minimize overhead time during scanning of the sample. The classification of detected events is automated and eliminates human reviewer error and permits true quantitative assessment of parameters. Imaging times of cells in a sample were reduced 3-fold by the development of cell alignment structures that concentrate cells onto the analysis surface. The automated classification of circulating tumor cells was further improved by the removal of contaminating unbound magnetic particles from processed patient samples. This also allowed for the accurate detection of treatment targets on cells of interest.Specialties(Cell) Biophysics, working with / building microscopes, sample preparation, image processing and analysis.
