Structural Biologist - Postdoctoral Research FellowFred Hutch May 2017 - Mar 2019Seattle, Wa, UsAt the Fred Hutchinson Cancer Research Center, I was working towards the development of a safe and effective vaccine against HIV-1. Our major emphasis was the design of protein immunogens and understanding the immune response following vaccination. My responsibilities involved the development and optimization of purification schemes for antibodies of interest and structural characterization of the interaction between antibody and antigen. Using molecular biology techniques, protein engineering, and high-throughput crystallography, I was able to solve multiple high-resolution crystal structures of antibody-antigen complexes. This work elucidated the structural mechanisms by which our antibodies were able to achieve strong neutralization phenotypes, and/or display breadth across different HIV-1 clades by targeting conserved regions in the HIV Envelope protein. I also worked collaboratively to characterize antibodies targeting Malaria MSP-1, and solved a crystal structure of an antibody bound to nicotine as a proof of concept for using vaccination as method to treat substance abuse. -

This approach provided insight into how the antagonistic relationship between these two families of proteins contributes to either cell cycle arrest or the progression of tumorigenesis. From this experience, I have gained familiarity with optimizing human protein and enzyme construct design, protein expression and purification methods to facilitate assay development, and structural characterization of a variety of protein complexes. The combination of structural and biochemical data with phylogenetic analyses provides novel insights into the coevolution of a protein–protein interaction critical for control of cell proliferation. -
