Woese Institute for Genomic Biology (IGB)Energy BioScience Institute (EBI)Prof. Yong-Su Jin & Dr. Soo Rin Kim- Metabolic Engineering (EBI)Elucidated the underlying mechanism for efficient cellulosic bioethanol production in S. cerevisiae expressing a heterologous xylose pathway. This was done by perturbing the genome of S. cerevisiae strains in order to understand the mechanism and role of a HAD phosphatase, Pho13 enzyme, through assessment of gene expression levels by various RNA quantification techniques, incubations, and assays. Assessment included investigating global transcriptional changes as a result of Pho13 deletion and the observation of the resulting oxidative stress response and phenotype. -

Hergenrother- Drug Discovery Focused on the development of novel small molecules to inhibit complexation between MBNL1 splicing regulator proteins and CUG RNA repeats to treat myotonic dystrophy type I (DM1). In order to do so I designed a low-cost and time efficient electrophoretic mobility shift assay which was able to preliminarily screen small molecules that disrupt complexation between disease specific RNA and MBNL. Using the developed assay, the toxicity of previously selected and designed compounds was determined by culturing HeLa cell plates as well as disease specific DM1 patient cell lines in order to test compound viability in vivo. Assessed effectiveness of selected compounds by visualizing disruption of the RNA-MBNL complexation through disruption of fluorescent foci in cell culture by confocal microscopy. -

The scaffold was made using an online software (Freiburg RNA Tools) and through communication with a doctoral student under Prof. Pamela Silver (Harvard University). Proved scaffold function through EMSA using site specific RNA binding proteins tethered to non-specific endonucleases through collaboration with Prof. Zefeng Wang (UNC-Chapel Hill) and further characterized the RNA scaffold through AFM imaging. -
