Research resulted in the development of new enzymatic mechanism for B. sub oxalate decarboxylase that identified the cleavage of the carbon-carbon bond through the redox cycling of its manganese cofactor that is facilitated by the controlled formation of superoxide during reaction.♦ Designed and developed flow cells for the use with real time kinetic measurements of free radical production. EPR spectrometer was interfaced with an HPLC for front end separation prior to collection of EPR spectra and mass spectrometry (LC/EPR/MS). Resulted in definitive confirmation of controlled superoxide and carbon dioxide radical formation of B. sub oxalate decarboxylase.♦ Identified Mn(II)/(III) redox cycling and molecular geometry elucidating coordination of metal cofactors in B. sub oxalate decarboxylase by sub-kelvin X-Band EPR (UF) and multi-frequency EPR investigations utilizing a multi-frequency/field quasi-optical broadband spectrometer at the National HighMagnetic Field Laboratory (Tallahassee, FL).♦ Solved the first crystal structure of the active pH structure of B. sub oxalate decarboxylase (in preparation for publication). ♦ Designed and developed an immobilization protocol for affinity tagged metalloenzymes for use with low temperature EPR. System is amenable to any protein sample with a His-6X affinity tag and a paramagnetic probe. ♦ Characterized and definitively identified formation and localization of hot holes and electron movement across rutile and anatase phase Ti-Oxide and Au-coated Ti-Oxide materials. Characterized photo-efficiency and viability of material to act as solar cell under visible light conditions.♦ Identified spectroscopically the formation and migration of polarons and bipolarons in true black polymer films developed for use in displays. Cyclic voltammetry conducted in concert with EPR identified switching voltage for color transition.
