Protein pH Stability• Study the notion of “Stability promotes evolvability”. Construction of a random mutational library of highly kinetically stable protein variant by a directed evolution approach. •Development of high-throughput screening method to identify pH stable protein variants.Multifunctional Protein Assembly and their synergistic effects • Engineered proteins with a charge peptide sequence to study the self-assembly of multifunctional enzymes and their functional ability. Protein Thermodynamic and Kinetic Stability• Applied a recombinant DNA technology and a novel transposon method to construct insertional gene fusion library. • Developed a stringent screening method to identify the most stable protein variants from the library by eliminating 90% of the non-functional/non-stable variants.• Employed CD spectroscopy, DSC thermoanalytical tool and other colorimetric techniques to characterize the kinetic and thermodynamic stability of selected protein variants. DNA Transposons and Novel Methods• Design and engineered “CP- ISC” and “CP- ISSC” transposons and developed a novel method for the construction of random circular permutation libraries of a protein. • Design and engineered “MuST” transposon and developed a novel method for easy construction of random domain insertion libraries of proteins.• This development of new transposons and transposition methodology creates randomly fused and circularly permuted proteins by means of DNA editing.
