In Vitro Toxicology Laboratory

I worked with the In Vitro Toxicology laboratory as part of my final year project under Dr Gillian Conway’s and Prof. Shareen Doak’s supervision. My final year project is titled “The Genotoxic and Pro-inflammatory Impact on 3D HepG2 Spheroids Following Carbon Black and Multi-walled Carbon Nanotubes Treatment”. Key responsibilities:• Maintenance of cell line (feeding and sub-culturing)• Good laboratory practice (correct labelling)• Developing spheroids by the hanging drop formation method with agarose• Cytokinesis-block proliferation index (CBPI) counts and Trypan blue counts • Developing cytodots and fixing slides • Micronucleus (MN) Assay • Using the Metafer to score slides • ELISA for pro-inflammatory markers (IL-8)• Ensuring research is conducted in a timely manner • Organisation of myself, research, and others

Swansea Worm Integrative Research Laboratory

This was a paid summer internship looking at the use of L. variegatus as a teaching model for in vivo pharmacology with Dr Aiden Seeley. We examined the effects of nicotine on L. variegatus behaviour when administered with drugs commonly used for nicotine cessation (tubocurarine and mecamylamine). The aim of this project was to determine whether the effects of nicotine on L. variegatus could be augmented by co-administration of these drugs to further understand the fundamental basis of addiction. Key responsibilities:• Maintenance of the worms (feeding and cleaning the tank)• Free locomotion assay • Stereotypical movements assay• Protein extraction and western blot to identify nicotinic acetylcholine receptor homologs within this worm species.• Analysis of Z-stacks on Image J
