Use of the techniques of molecular biology to clone, express, and purify MGH as a model recombinant protein. Determine the key kinetic constants/parameters of MGH using appropriate equations of enzyme assays. This lab consists of 3 sections with a total 36 students.Responsibilities-• Written and revised the graduate assistant manual for each experiment.• Prepare the inventory for laboratory chemicals and materials at the beginning of every semester. Communicate with lab instructors, and stock in charge for approvals. After approval communicate with the lab director for final purchase.• Assist undergraduate students in addressing inquiries and providing assistance.• Calculate concentrations of required chemicals and Prepare chemical solutions, buffers, bacterial cultures, LB media, agar plates, aliquoted bacterial cells (DH5 alfa,BL21 DE3 PLySs), and prepare the master mix for the PCR reaction, and enzyme solutions.• To start the overnight cultures for each section, make available materials, chemicals, and equipment.• Autoclave, and sterilize the required material for experiments.•Cleaning the lab and disposing of the Biohazardous materials with the CNSM Biosafety officer. Protein quantification and analysis done by using SDS-PAGE and Western blot analysis.• Nanodisc prepared- The proteins were reconstituted with phospholipids followed by density gradient ultracentrifugation to isolate lipoprotein particles. • Characterized enzyme containing nanodiscs using multiple biochemical methods such as colorimetric assays (PL/BCA assay), Western blot and physicochemical characterization using native PAGE and co-immunoprecipitation.• Assessed enzyme activity by sphingomyelinase activity assay kit.• Cellular uptake and lysosomal localization were determined through fluorescence microscopy and confocal microscopy.• Determination of structural characterization of the chimera through fluorescence and circular dichroism spectroscopy and flow cytometry are in progress.• Utilized problem-solving skills to optimize experiments to complete objectives• Designed, implemented, and evaluated data from experiments to achieve research objectives and meet deadlines.• Data optimized and presented to research mentor and local, and national research conferences.
