My laboratory is focused on neurodegenerative diseases and conditions affecting the retinal function such as glaucoma, diabetic retinopathy, age-related macular degeneration and Alzheimer's disease. Our main objective is to design treatments to rescue retinal neurons from cell death and stimulate their recovery in ocular pathologies using pharmacological compounds, neutralizing antibodies, gene therapy and nanoparticles in animal models. The effects of new therapeutic approaches are tested by electroretinography, with behaviour tests and by retinal imaging in living animals. Classical histological, cellular and molecular biology techniques are routinely used to monitor the effect of treatments on the course of degenerative processes in the retina and in the visual brain.

As a Research Associate in the laboratory of Prof Martin E Schwab at the Brain Research Institute in Zürich, I established a group dedicated to research in neuro-ophthalmology and composed of 5 graduate students, 1 postdoctoral fellow and 1 technician. Our team made important contributions to the fields of developmental angiogenesis, axonal regeneration, glial cell development, neuroprotection, retinal stroke, and visual projection plasticity in the brain of mice. State-of-the-art techniques were undertaken to control the activity of proteins of interest, for instance by virus-mediated gene transfer (adeno-associated virus, adenovirus, lentivirus), immunotherapy, pharmacology or by genetic KO/KD.I obtained individual research grants from the Velux Stiftung foundation and the Universität Zürich (Forschungskredit).I attended international meetings organized by the Association for Research in Vision and Ophthalmology (ARVO) and the Society for Neuroscience (SFN).

I conducted experimental projects consisting of explorating new functions of a CNS myelin protein, Nogo-A, using the mouse visual system as a model in vivo. After traumatic lesion in the optic nerve, different methods were applied to monitor molecular changes occuring in the retina (qRT-PCR, Western blot, immunofluorescence) and to follow retinal cell survival (e.g. confocal imaging). Axonal tracing allowed to visualize the growth of injured optic nerve axons by confocal microscopy. Later on, a new 3D imaging was developed to observe the course of regenerating axons at an unprecedented level of resolution. These results are of great importance to understand the molecular mechanisms involved in CNS axon regeneration and may have major implications in the treatments of injuries in other regions such as the spinal cord and the brain.
