The enhanced sensitivity can facilitate the shift of existing lab tests into point-of-care testing, and enable the use of non-invasive biomarkers like saliva. Dsyregulated receptor tyrosine kinase (RTK) signaling is commonly associated in cancer development and progression. The goal of the laboratory headed by Axel Ullrich is to elucidate novel RTK pathways driving carcinogenesis and identify potential targets for developing small molecule kinase inhibitors. My project focused on elucidating the role of klotho-beta (KLB), a newly discovered membrane co-receptor of FGFR4 in hepatocellular carcinoma (HCC) progression. Given the high recurrence and low five-year survival rate of this disease, this research aimed to shine light onto new pathways for therapeutic intervention. The research involved sequencing and quantitative real-time PCR analyses of paired liver samples from patients to uncover mutations, abnormal expression of KLB in HCC, and subsequently correlating this information with clinicopathological features. We showed that KLB gene expression is significantly upregulated in tumor liver tissue relative to their paired normal tissues and silencing KLB expression decreased cell viability of HCC cell lines. Future work will involve studying novel downstream targets of KLB and developing small molecule inhibitors to target the KLB-FGFR4 signaling pathway in HCC tissue.
