About Weiming Ni

● Accomplished research scientist in the fields of protein folding, neurodegenerative disease, metabolism, vascular biology and immunology with 16+year-experience of research. Published more than 10 research and review articles in world’s leading peer-reviewed biomedical journals including Circulation Research, PNAS, Nature Communications, Oncogene, Journal of Neuroscience, Journal of Cell Science and etc.● Established transgenic C. elegans for chaperone study, generated stable SOD1 oligomers for neurodegenerative toxicity investigation in different models, explored O-GlcNAc protein interactome via BioID, and utilized tight junction formation to study clinical vascular leakage. Have employed single-cell technology to study various types of cells’ cytokine secretion, intracellullar proteins and etc. Have collaborated inside/outside to study squid axon transport, electrophysiology, RNA folding, mouse food intake, oncogene, CRISPR and etc. ● Extensive hands-on experience with the following techniques and activities: Primary Cells isolation and cultures (endothelial cells), ECIS (electric cell-substrate impedance sensing) and transwell (FITC-dextran)Microinjection (Aplysia bag neuron and c. elegans gonad), immunostaining (cell and human/mouse tissue)Protein purification, Mono Q chromatography, FPLC, western blot, pulldown, IP (immunoprecipitation), protein interactome Activation and inflammatory cytokines (TNF-α, IL6, LIF, OSM and etc) expression assay RT-PCR, qPCR, RNA isolation, transfection, ELISA and etc● Strong communication and documentation skills; an outstanding ability to work effectively in interdisciplinary fields and to manage multiple projects.

    • Scientific leadership: advance new assays for analysis of immune cellular reactions to various stimuli on IsoPlexis’ system• Create methods from sample preparation to data analysis in various disease models to analyze single cellular reactions to therapy• Design immune cell experiments, conduct routine analysis, interpret large data sets using statistical methods and visual tools• Manage and mentor scientific staff to advance projects• Report analysis for potential biomarkers of patient response and discuss with collaborators• Engage in theoretical discussions around strategic scientific developments across projects• Understand the latest literature and lab technologies to inform novel methodologies for all assay development as they apply to pre-clinical programs for biopharmaceutical products
  • Yale University School Of Medicine
    Associate Research Scientist
Yale University School Of Medicine Sep 2016 - Feb 2021
    New Haven, Ct, Us
    1. Employing endothelia cells as a model to explore vascular biology therapy in combination of clinical data. Using ECIS (electric cell-substrate impedance sensing), transwell (FITC-dextran), RNAseq, shRNA, siRNA, WB, immunostaining and etc to find the molecules/mechanism for rescuing vascular leakage.2. Have studied metabolism-related diseases. Utilized BioID to investigate O-GlcNAc protein interactome via knockin of BirA into 293 cells
  • Howard Hughes Medical Institute/Yale Univerity School Of Medicine
    Postdoctoral Fellow
Howard Hughes Medical Institute/Yale Univerity School Of Medicine Dec 2011 - Sep 2016
Studied neurodegenerative disease with in vitro and in vivo approaches. 1. Made stable SOD1 oligomers via Talon resin, monoQ and FPLC after expression in BL21. Injected oligomers into Aplysia bag neurons and then checked current/voltage changes. Also injected siRNA/pharmacological chemicals to look for the channel/kinase which are in charge of the electrophysiological changes.2. Used a new setup of electrophysiological device to monitor SOD1-linked ALS mice for disease progression while walking on the wheel.
