One possible solution is to culture tissues from induced pluripotent stem cells (iPSCs). These stem cells have the potential to form any type of tissue in the body and these tissues can be used to support organs and to restore their function. However, these tissues can be rejected by the recipient's immune system after transplantation.To prevent tissue from being seen as foreign, we can genetically modify the cells. With the help of CRISPR-Cas9, very specific changes can be made in the DNA that ensure that the cells are protected against the immune system of the recipient.In this research we want to investigate both in vitro and in vivo which specific modification is most effective in protecting the tissues against the immune system. To allow production of iPSC-derived clinical products the iPSC lines need to be produced under GMP compliant conditions. LUMC is currently translating lab-based practice to the LUMC GMP facility. This entails writing of GMP standard operating procedures (SOPs) and the design of release assays. The research group makes use of episomal plasmid for somatic cell reprogramming. The hiPSC when released for clinical production of differentiated products should be free from episomal plasmids. “Absence” of plasmids in the cell products is determined by quantitative polymerase chain reaction (qPCR).
