About William Herrick

I am a scientist in the Clinical Pharmacodynamics Biomarker Program at the Frederick National Laboratory, where I develop novel, quantitative sandwich immunoassays on the Luminex platform to support pharmacodynamics research in NCI-sponsored clinical trials. My interests are in the development and validation of assays and antibodies, the pharmacodynamics of molecularly-targeted therapies, and the development of industrial bioprocesses (e.g. cell culture processes).

    I am a postdoc fellow in the Pharmacodynamics Assay Development & Implementation Section (PADIS) at the Frederick National Laboratory (NCI-Frederick) conducting Luminex multiplex assay development in the area of MEK/ERK and PI3K/AKT signaling. The position is challenging but also rewarding, as it provides an inside-look at cancer drug development and clinical trial design at the NCI. I am also excited that my assays will someday be used to aid in the development of more effective drugs and therapies by providing critical pharmacodynamic information and drug resistance mechanisms for NCI-sponsored clinical trials.My primary responsibilities include monoclonal antibody development to generate highly specific antibody reagents and all aspects of Luminex assay development. Other critical duties include the screening and validation of commercial antibody reagents, generation of validation materials from in vitro cell culture models, selection of recombinant protein standards for assay calibrators, reagent scale-up, and assay fit-for-purpose validation. Details of major responsibilities:Monoclonal Antibody Development: - Design of immunizing peptides - Screening of animal test bleeds and hybridoma supernatants for desired specificity and optimal affinity - Antibody purification by Protein A/G affinity chromatography - Assessment of antibody candidates in Luminex assaysLuminex Assay Development: - Development and selection of antibody and recombinant protein reagents - Antibody coupling to Luminex bead reagents - Antibody biotinylation - Assay optimizations including buffer selection, detection and capture antibody titration, standard curve range, and sample preparation (with major considerations to pre-analytical variables) - Assay validation including assessment of spike-recovery, cross-reactivity, specificity, dilutional linearity, reagent stability, inter- and intra-assay precision

    During the time between finishing graduate school and starting my current postdoc position, I was a part-time after-school math instructor at the Mathnasium in Mendham, NJ. My role was to work with students (K-12) 1-on-1 in a group setting to teach them fundamental and practical concepts from numerical fluency and problem solving to more advanced math topics including geometry, trigonometry, pre-calculus, AP calculus, and SAT prep. Despite having little prior teaching experience I was able to quickly master the tools, strategies, and techniques needed to effectively teach students of all ages and skill levels. It was an extremely rewarding experience and I will miss it!

    With my original adviser, I engineered an enzyme with the long term goal of producing a quorum sensing yeast for autoinduced production of biopharmaceuticals: • Engineered a variant of the enzyme LuxI with directed evolution and selection to broaden substrate specificity.• Demonstrated with a cellular assay and HPLC-MC that our novel enzyme variant produced a wider variety of signaling molecules.• Functionalized this variant in yeast (S. cerevisiae) by inducing mitochondrial import with a genetically-encoded targeting sequence, required because mitochondria are related to bacteria and contain the necessary enzymatic substrates. • Verified that mitochondrial import was necessary and sufficient for activity in yeast by expressing a custom fusion protein in S. cerevisiae and confirming production of the signaling molecules with a cellular assay.After my original adviser relocated, I joined an innovative new lab, where we develop useful biomaterials and study how cell-microenvironment interactions impact disease: • Developed and characterized a novel PEG-based hydrogel with exceptional mechanical properties (~1 - 10,000 kPa Young's modulus), fouling properties, and optical transparency.• Applied this material to a mechanobiology study investigating the effects of substrate stiffness on proliferation and adhesion of multiple cancer cell types. (Herrick et al 2013)• Developed an in vitro model featuring multiple physical and chemical factors to investigate their roles in the phenotype plasticity of smooth muscle cells and disease-relevant behaviors including proliferation and motility. Requirements included extensive mammalian cell culture, numerous immuno (xMAP, Westerns, IHC/IF) and cellular (proliferation, viability) assays, microscopy, and more. (Herrick et al 2015)• Juggled multiple lab management duties including initial start-up, the training of new students, supply ordering, and the ordering, setup, and evaluation of new lab equipment.
