• Close collaboration with several Target Biology groups, Protein Biochemistry, and In Vivo Pharmacology for antibody discovery/characterization/lead candidate selection, protein needs, assay development, and target biology experiments• Successful humanization of all 11 antibodies attempted, discovery of lead or surrogate antibodies using phage display, affinity maturation of antibodies via phage display and targeted mutagenesis, and discovery of surrogate antibodies derived from rabbit hybridomas• Characterization of antibodies through kinetic analysis (Biacore, Octet, KinExA, FACS), biophysical studies, epitope binning/mapping and functional assays• Large/small scale protein production, including CHO stable cell line generation• Protein engineering, including the design and generation of antigens as well as Fc fusions as potential therapeutics• Blood-based biomarker discovery for multiple programs• Assay development, including neutralization of ligand/receptor binding, pharmacokinetic, and cell-based• Target validation and investigation of target biology• Coordinated large effort for new target nomination using bioinformatics and literature based approaches• Development and validation of antibody engineering/discovery technology• Extensive use/programming of liquid handling robot (Tecan EVO-200) • Creation of inducible RNAi tools• Author of numerous patents, reports, and IND documents• Member of CMC teams responsible for the sourcing and management of contract manufacturing of antibodies• Member of teams responsible for the coordination of external antibody partnerships
